Dear Ross

The post I have advertised will follow on from our recent publications
showing SNARE protein association with lipid raft domains. I still have to
finalise exactly what portion of the work will be taken on by the RA
employed, but it is likely to focus on SNARE proteins that function in
Glut4 vesicle exocytosis (in fat cells).

The direction we are taking is to generate mutant SNAREs which have either
an increased or decreased affinity for lipid rafts and to assess the
effects of these mutants on exocytosis. We are also using mutagenesis to
understand how SNARE proteins are targeted to rafts.

A major portion of the work taken on by the RA will investigate the
differential localisation of VAMP2 and cellubrevin in rafts. In fat cells
we find that >70% of VAMP2 is raft associated but only a small postion
(15%) of cellubrevin is in rafts. We would like to know whether this
difference is due to specific differences in the amino acid sequences of
these vSNAREs or whether it reflects the differential localisation of these
proteins in fat cells (VAMP2 is mainly found in GLUT4 vesicles, callubrevin
in the endosomal system). Other work will look at the effects of decreasing
the raft association of SNAP23 on GLUT4 vesicle exocytosis.

Techniques that would be useful to have experience in would come from the
following: gradient centrifugation, cell fractionation, SDS-PAGE,
immunoblotting, immunofluorescence, mammalian cell culture and transfection.

I think there should be no problems registering for a PhD (although the RA
may have to pay fees- although there may be ways around this).

no i got few ideas atm which im working on , business wise
