in theory the 7200 rpm, but wont a massive amount in it.
The biggest bottle neck is from the platter rotation / cache and not the connecting interface.
If you want faster sustained speeds, go for 7200.

Go for 7200rpm if possible.
SATA II or III makes no difference, the physical limitations of a mechanical hard drive mean that you won't benefit from 3Gb/s or 6Gb/s transfer speeds.
Where this DOES matter though is if you buy a solid state drive (or SSD for short), as they have no mechanical parts.

ive already got a sata 3 ssd I'm just looking to increase the size of my secondary drive, i can only seem to find 5400 sata 3 or 7200 sata 2

Would go with the 5400 tbh.
Speed gains of the 7200 will be minimal compared to the impact on noise and battery life.
