no not glue, sealent mate...the stuff you wack around baths, but get flexible stuff, otherwise it will crack under the pressure from the subs. And im sure the box will be fine without, but while you have everything out i would just wack some it, it will cost about 3 quid for a tube of the stuff from Do It All, or B&Q etc 
As for wadding, yea its a personal preference....but have a go with it mate, you might like the sound with it in....once you have everything install, just take out one of the subs, wacks some wadding in, screw it back and have a listen for a fews days. If you dont like it, take it out mate
